Gelsolin is a Ca2+-sensitive, actin filament end-blocking protein which promotes actin nucleation and also severs actin filaments. It was first identified in rabbit lung macrophages I and subsequently found in many mammalian cells including those of muscle. A variant of gelsolin exists in blood plasma and is a bona fide secreted protein, z A number of protocols have been used to purify gelsolin from different sources. Before describing details of the purification, properties of gelsolin which are critical for designing a gelsolin purification scheme will be discussed.
